Echocardiographic technology not technique ; is introduced, emphasizing aspects that are not widely known to general practitioners, such as 2D, M-Mode, Color Doppler and Spectral Doppler imaging. Rather than teaching practitioners how to acquire echocardiographic data, the aim is to expose them to the types of information that can and should be gained from a well equipped echocardiographic examination performed by a thoroughly trained professional, to help them solve daily diagnostic and therapeutic dilemmas. At the end of the lecture they will be familiar with concepts that appear on a written echocardiographic examination report, such as received by a cardiologist in response to a cardiac referral. Echocardiography, like all other diagnostic technologies, * cannot * and should not substitute an effective physical examination. Rather, it should help us confirm or rule out diseases we diagnose or suspect, after having prioritized their likelihood based on the 4 simplest and most important bases of the diagnost process: signalment, history, observation, and physical examination. Most veterinary cardiac patients especially cats, puppies, and many adult dogs ; are either too unstable or too impatient to allow a lengthy echocardiographic examination. For each of these, an overview is given of current production technologies, of their properties, the technical substitution potential, the production cost and the environmental impacts. The order followed in this chapter roughly represents the current importance of each group of bio-based polymers in terms of production volumes in Europe see Tables 2-1 and 22 ; . Starch polymers and polylactic acid PLA ; are now clearly the most important types of polymers. Aliquots of the amplification product 5 l ; were analysed by gel electrophoresis in 1% w v agarose containing ethidium bromide 0.5 g ml ; . DGGE was performed using an Ingeny PhorU-2 system. 10 L of PCR samples were applied directly onto an 8% w v polyacrylamide gel in 0.5 x TAE 20 mM Trisacetate pH 7.4 ; , 10 mM acetate and 0.5 mM Na2EDTA ; . The gradient was formed with 8% w v acrylamide stock solutions acrylamide N, N'-methylene bisacrylamide, 37: 1, w w ; containing 20% and 80% denaturants 100% denaturant being 7 M urea and 40% v v formamide ; . Electrophoresis was performed at a constant voltage of 100 V and a temperature of 60C for 16 h. Following electrophoresis, the gel was stained for 1 h in Milli-Q water containing 0.5 g ml ethidium bromide and destained in Milli-Q water for 75 min. The gel was then visualised and photographed under UV 302 nm ; transillumination Kodak Digital Science v1D ; . Bands of interest were excised and the gel pieces soaked in 20 l Milli-Q water for 4 h at 4C. PCR was performed with Lac1 and Lac2GC primers as previously described, using 2 l of the gel extract solution as DNA template. Amplicons were verified by gel electrophoresis. A 10 l sequence mix was then prepared using 2 l of the resultant PCR product, 1 l of a 3.2 pmol l Lac1 primer solution, 3 l MilliQ water and 4 l BigDye v3.1 terminator mix Applied Biosystems, UK ; . The sequencing reaction was performed as follows: 25 cycles of 96C for 10 s, 50C for 5 s and 60C for 4 min. Products were sequenced via an ABI Prism 3700 capillary sequencer Applied Biosystems, UK ; . Sequences retrieved were of a minimum length of 320-bp data not shown ; . Strains were identified by comparison with known sequences obtained from GenBank and EBI databases via a BLAST search Maidak et al., 1997 ; at 99.5% sequence homology. The State alleged that the defendants had violated the Act by falsely advertising the capabilities of the dust masks they sold, which are used in industrial settings, such as coal mines. Specifically, the State alleged that while the dust masks were marketed as being safe and effective, the masks failed repeatedly, resulting in the State incurring larger than anticipated worker compensation claims and other health related costs and expenses.
